この地域には、大伝馬町の分名主で、公用旅行者のための人馬の継立てなどを行う道中伝馬役を務めた馬込氏の屋敷がありました。 馬込氏は、天正十八年(一五九〇) に徳川家康の江戸入府に際して、高野新右衛門・小宮善右衛門らとともに取馬人足を率いて出迎えたことにより、道中伝馬役を命ぜられて、後の呉服橋御門 (千代田区丸の内一丁目) 内の辺りに位置した宝田村に土地を与えられました。馬込氏の当主の多くは、代々「物解由」の通称名を名乗っていました。 慶長十一年(一六〇六) 江戸城拡張にともない、宝田村の住人たちが現在地付近に移転をして、大伝馬町が起立しました。大伝馬町は、京橋にある南伝馬町と交代で道中の人馬の継立てを行い、江戸府内の継立てを務める小伝馬町と合わせて「三伝馬町」と呼ばれました。大伝馬町の伝馬役を務めた馬込氏は、大伝馬町二丁目北側新道の西角に屋敷を構え、同町の名主役を兼帯して苗字帯刀を許されていました。 馬込氏の邸内奥には、宝田村の鎮守が勧請され、その御神体である恵比寿神像は徳川家康から拝領したと伝えられています。正月二十日と十月二十日には大伝馬町の大店で恵比寿講が盛大に行われ、その前日に開かれた市が、現在十月に開かれている「べったら市」につながっています。なお、馬込氏邸内の恵比寿神像は、江戸時代後期頃から現在の身田恵比壽神社 (日本橋本町三丁目十番十一号)にまつられています。
